Who says they do? Assassination is obviously a secretive thing but the most well known assassins have been military teams or ex-military. Obviously they’d learn most of their unarmed combat in the military! Then there are the “civilian service / clandestine service” assassins that aren’t even supposed to exist. They, too, would probably learn from military guys or while they were in the military. In WW2 there was Camp X-Ray for OSS which was technically military but sort of also not military…?
But don’t forget the whole point of an assassin (in the modern, kill a public figure, sense) is to murder. Not to fight. If they have to fight they’ve probably done something wrong. Look at the famous assassinations and attempts we know about. They’re mostly sniper attacks, bombings, pushing someone in front of a bus, poisoning, machine-gun blitzes, ambushes, etc.
Even ninja (real term Shinobi-mono) despite their fearsome reputations weren’t expected to fight but to kill and their assassinations usually used very unorthodox methods such as disguising themselves as women or monks or pilgrims and getting within firearms range, poison darts and so on. In one famous case a ninja hid in the toilet of a castle until the lord went to poop and then shoved his spear up where the sun don’t shine. Can’t imagine there was a lot of boxing going on after that!
And then let us not forget that modern assassination often takes completely different forms. I still remember the first night of the Iraq war when stealth bombers turned Saddam Hussein’s residence into a cloud of flaming dust. We can call that whatever we want, it was a straight-up assassination attempt. I’m not saying it wasn’t justified, that son of a bitch had to go and it would have saved a lot of lives if he’d have met his fate that night, but let’s be honest about what it was.
And in the case of Cartel types? I’m going to guess that if they learn they learn it growing up in some hell hole. Life is not easy in a drug ghetto.

The importance of hand to hand combat has been overstated by movies, comic books and video games. In the real world i.e. the streets or the battlefield, hand-to-hand combat is rare and for good reason. A 10-year old with an uzi or 10 armed men with little training can do more damage quickly than one 7th degree black belt in the deadliest martial art. Movies use the ‘one-against-many’ combat scenario for thrills but its just choreography. Numbers and size matter in the real world.
Assassins are trained to be covert. They utilize social engineering and deception to get close to the target and execute them. After that they are prepared to be expended. Hand-to-hand combat is really not that important.

Same place anyone else. For government and organisational based ones, by their designated CQC trainer, for a civilian one, down the gym. There’s no ‘secret techniques’ here, and they’re unlikely to even be that good, no assassin is going to be punching his target.
What an assassin needs is escape techniques, which he will likely be good at. Slipping out of holds, keeping space, throws and shoves. He’s not going to want to get into a stand up fight with anyone.

Besides the usual—the military, martial arts, combat sports, groups in prison that practice killing techniques, and just other people in their trade or related criminal trades—I thought it was interesting that some benefit from experience with animal husbandry/butchery.
I have heard of secret “schools” where killing techniques are passed along from one criminal to another but assume they are rare. Nigel February of South Africa says he has heard of such a place but he was not speaking in the context of assassination specifically.

I would guess you would be better off with a pharmacology degree in a low-conflict situation and military/weapons training in a high-conflict one.
Now when it comes to non-lethal small-scale enforcers most seem to come with a regular martial background, usually a no-nonsense full contact sport ( specifically (kick)boxing and judo in cases that I personally know of).
T raining is a long process, two-months courses into superhuman killing machines is for movies, not RL. Recruiting people that have both an existing martial background and street experience is by far the most reasonable way to go.

I imagine guile and craft would be used more in either attacking your target while being totally unaware of your presence and intentions. Real easy to analyze open vital areas while your target is on the move or attention focused elsewhere.

Assassins make a living killing people, not fighting with them. Some are trained, just like our military, police and a whole lot of regular Joes on the street, in hand to hand combat to varying degrees, some are not. So, those that do learn just like other folks do, from people that know and are skilled in hand to hand combat technique and strategy and from experience.
