If the combat knife you are referring to is the one on your Spartans chest or so, I have to inform you that they are purely cosmetic. If you’re referring to the insta-kill move you’ve seen people do that involves a knife, you need to be behind a enemy and hold the melee button slightly. The exception is hunters, crawlers, any enemy that occupies a vehicle, if you are in a vehicle and multiplayer games where assassinations have been disabled.

In all Halo games (campaign and multiplayer) since Halo: Reach, you need to be directly behind an enemy in order to use your knife. When you are in position, press and hold the melee button, and you will perform an assassination.
The assassination that you perform depends on what you’re doing at the time (running, jumping, walking, falling, etc.). Not all assassinations involve you taking out your knife, but many of them do, and all assassinations are instant kills.
There are a few exceptions though: First, you cannot assassinate Hunters, at all. Second, in order to assassinate a Promethean Knight, the Knight cannot be aware of you at the time— that means you need to sneak up on it silently, without it knowing you’re there. Because of that, opportunities to assassinate a Knight are pretty rare. It’s pretty tricky to assassinate a Knight, but I’ve found it works best when the Knight is distracted and busy fighting some other enemy.

That is because there is no combat knife (or at least none that you can pull out).
All regular melee’s are essentially just bashes with your gun or otherwise.
HOWEVER, you can use a knife of sorts when you assassinate someone either in multiplayer or in the campaign mode.
What you have to do is hold down the melee button when you are directly behind someone or behind them slightly to the side. Front facing either to the sides or directly will not trigger the assassination.
Another thing to note certain in game units cannot be assassinated such as the Grunts and the Hunters I think. Otherwise everything else is pretty much the same.
